Abstract
Random amplified polymorphic DNA (RAPD) markers were used to identify the interspecific hybrids of pepper crosses: C annuum x C. chinense and C annuum x C baccalum and to identify the possible markers to tag disease resistance genes from the wild species (C. chinense and C. baccalum) on the hybrids. Forty Operon primers were screened to detect for polymorphism among the 12 parents used in the study. Twenty-six primers were found useful in screening the parentals, 12 of which were used to test the progenies. Of the 12 primers used for hybridity testing, 7 primers (OPAN3, OPAN6, OPAN8, OPAN9, OPANI I, OPAN15, and OPAE7) identified the interspecific hybrids. The primers OPAN3, OPAN6, OPAN8, OPAN11, OPAN15 and OPAE7 identified the interspecific hybrids of the cross C. annum (PBC535, PBC950, PBC972, PBC204, PBC7I5, PBC122 11 and PBC534) x C. chinense (PBC932 and PBC323). Results showed that for the cross of PBC534 x PBC932, the progenies assigned with DNA numbers 200, 201, 205, 210 and 214, using OPAN3 primer are not hybrids because of the failure of both parental bands to appear on the progenies. Primers OPAN3 and OPAN9 identified the interspecific hybrid of the cross C. annuum (PBC534, CCA187, PBC204, PBC950, and PBC535) x C. baccalum (PBC81). OPAN3 identified the hybrid of CCA187 (hybrid of the cross of PBC122 x PBC932) x PBC81. Those primers that identified the interspecific hybrids needs further analysis for disease resistance in the F2 generation. The study reveals that RAPD markers can identify possible markers to tag for disease resistance genes coming from the wild parents.
