Abstract

Total globulins were extracted from mature coconut (Cocos nucifera L.) endosperm using 0.4 M NaCI. Gel filtration chromatography of the globulins resulted in the isolation and purification of two peaks. Peak I was cocosin, the 11S globulin in coconut. Peak 11 was the 7S globulin. SDS PAGE of the peak LI fractions revealed four bands: 55 k, 24 k, 22 k and 16 k. Fractions containing the 7S globulin were dialyzed without f3-mercaptoethanol to detect presence of disulfide linkage. SDS PAGE of these fractions showed no changes in the banding pattern of the protein. Thus, no disulfide linkages exist in the 7S globulin. The globulins were sequentially treated with increasing concentration of NaCI to test solubility of the protein. The 7S globulin was most soluble in the 0.15 M salt extract. The results indicate that the 7S globulin can be isolated and purified on the basis of its solubility in 0.15 M NaCl. Lastly, the 7S globulin tested negative for Con A-1-IRP staining and the periodic acid Schiffs staining for carbohydrates, indicating the absence of carbohydrate moiety.
