Morphological and cytological evaluation of partially sterile and fertile anther-derived doubled haploid lines from IR 64

Abstract
A study was conducted to determine the agro-morphological and cytogenetic characteristics of fertile and partially sterile sister doubled haploid lines (3 pairs), randomly taken fertile doubled haploid lines and their parents, IR64 and Azucena. Parents differed in thirteen characters, randomly taken doubled haploid lines in thirteen, sister line SM 128F and SM 128S in six, sister line SM 112F and SM 112S in eight and sister line SM 107F and SM 107S in one. Variability between sister lines could be due to out-crossing or mutations during anther culture. All entries had low pollen and spikelet fertility. This could be due to drought experienced by the crop under upland condition. It was noted that lines that were classified as fertile tended to have higher pollen and spikelet fertility than their respective partially sterile sister lines. Cytological analysis revealed a wide variation in chromosome number for all entries. Chromosome behavior was generally the same for all entries in terms of early disjunction, presence of non-congressed chromosomes and chromosome groupings at metaphase I. Low pollen and spikelet fertility noted could be partly due to high frequency of aneuploid cells which were potential sources of non-functional gametes and partly due to physiological disturbances induced by drought. Minimal differences in cytological and morphological characters were observed between sister lines, but great differences between non-sister lines and between parents. These results thus indicate that there is no direct association between chromosomal aberration exhibited by certain doubled haploid lines and the level of fertility It is further suggested that sterility observed in anther derived doubled haploid lines may be due to genic interaction.
