Abstract
The genetic variability within and between the natural populations of Apis cerena F. from Albay and Quezon were compared by examining nine colonies from each population using horizontal starch-gel electrophoresis. The frequency of occurence of each allozyme at different loci. namely, esterase (EST). malate dehydrogenase (Mdh), alkaline phosphates (ALPH) and isocitrate dehydrogenase (Idh) were determined. Est and Mdh exhibited variability in both populations while AIph exhibited monomorphism. Idh was polymorphic in Albay but monomorphic in Quezon. At the Est locus, the phenotypes EstS, EstM and EstF were observed with gene frequencies of 0.448, 0.322 and 0.230, respectively in Albay and 0.543, 0.435 and 0.024, respectively in Quezon. It was assumed that esterase is controlled by a single locus with three alleles. Mdh gene frequencies were 0.150 and 0.850 in Albay and 0.674 and 0.326 in Quezon for MdhS and MdhM, respectively. Idh had gene frequencies of 0.137 and 0.864 respectively for IdhS and IdhF in Albay and 1.0 for both phenotypes in Quezon. The goodness-of-fit was tested using the chi-square method and it was found that the observed genotype proportions for both populations do not fit the expected proportions according to the Hardy-Weinberg equilibrium. Between the two populations, the genetic identity value was 0.549 and the genetic distance was estimated at 0.260. The values indicate that there were significant differences between populations and presume that original colonies from each population are entirely different from the other.
