Enzyme variability in populations of the Asiatic corn borer, Ostrinia furnacalis (Guenee) (Lepidoptera: Pyralidae), from Camarines Sur, Isabela, and Laguna

Abstract

Genetic variability in populatins of Asiatic corn borer, (Ostrinia furnacalis (Guenee) from Camarines sur, Isabela and Laguna was determined through starch gel electrophoresis. Twenty larvae from Laguna were used in the study. while there were twenty-two from Camarines sur and Isabela. Ten presumptive loci encoding for four enzymes were observed. Three loci were observed for acid phosphatase (ACP), two for alkaline phosphatase (ALP), one for malic enzyme (ME), and four for esterase (EST_. ACP-1, ALP-1, ALP-2, ME-1, EST-2, EST-3, and EST-4 were each code for by two alleles, slow (S) and fast (F). Three alleles, S. moderate (M), and F, controlled EST-1; while only S allele coded for ACP-3. Only EST-3 were polymorphic for all three populations, on the other hand was monomorphic, occurring only in the Isabella population ALP-2 was absent in the Laguna population. The Isabela population was the most diverse based on the proportion of polymorphic loci (P=0.80), average number of alleles per locus (A=1.90) and average heterozygozity (H=0.33). the highest degree of genetic identity was observed between Camarines Sur and Isabela populations (0.813) while lowest between Camarines Sur and Laguna populations (0.602). Genetic distance was highest between Camarines Sur and Laguna populations (0.508) while lowest between Camarines sur and Isabela populations (0.207). These were supported by the dendogram produced using the NTSYSpc version 2.0. There were nine clusters at 0.70similarity coefficient, with the Laguna population forming two distinct clusters.
