ABSTRACT
Isozyme analysis on twenty five accessions of pili, Canarium ovation Engl. (originally from Oas, Albay) was conducted using starch gel electrophoresis. The plants were part of the gennplasm collection of the National Plant Genetic Resources Laboratory, Institute of Plant Breeding, UP Los Banos. The four enzymes studied were malate dehydrogenase (MDH), alkaline phosphatase (ALPH), acid phosphatase (ACPH) and isocitrate dehydrogenase (ICD). A total of 6 loci was observed, all of which are polymorphic except for ALPH-2. Two loci, MDH-1 with genotypes FF and FS and MDH-2 with genotypes FF and FS, and 5 banding patterns were observed in MDH. Alkaline phosphatase has two zones of activity, ALPH-1 with genotypes FF and FS and ALPH-2 with genotype FF, and 4 banding patterns. Two loci, ACPH-1 (genotypes FF, FS and SS) and ACPH-2 (genotypes FF, FM, FS and MM) and 8 banding patterns were observed for ACPH. No zone of enzymatic activity was detected for ICD. The degree of polymorphism and variability was evaluated. Out of the six presumptive loci observed encoding for the three enzymes, polymorphism was observed in five loci. The average degree of polymorphism (P = 0.8333) is high. The average number of alleles per locus (A) is 2.0. Five genotypes were each found to be shared by two accessions. On the other hand, the remaining 15 accessions exhibited different genotypes. These suggest variation in the accessions of pill trees used in this study.
