Dela Vina ABSTRACT Cytogenetic analysis was conducted in Carnoy's fluid-fixed specimens of male rice black bugs (RBB) Scotinophara coarctata Fabricius sampled from Dilasag, Aurora, Philippines. Slide preparations were made by first separating the abdomen of the males containing the testes and then macerating it in lacto-aceto-orcein. Slides were viewed under high power objective (400x) and photomicrographs were taken. All the stages of the first and second meiotic divisions were observed, as well as the interphase of the spermatocytes. The most encountered meiotic stage was Prophase I at 46.3%, while the least observed stage was Anaphase 11 at 0.1%. The average meiotic index of the species was 91.03%. At Diakinesis, the haploid chromosome number was revealed as n=8. The sex chromosome X was observed as a positively heteropycnotic body throughout prophase. Hence, the karyotype formula for the heterogametic male Scotinophara sp. is 2n=14 Autosomes + X. Photomicrographs of Pachynema and Diakinesis cells were used to measure the actual and relative lengths of the chromosomes, and a karyotype was constructed based on the said data. The average actual relative lengths of the Pachytene chromosomes ranged from 2.53 to 5.85 pm, while the average relative length ranged from 0.07 to 0.19 pm. Diakinesis chromosomes had average actual lengths that ranged from 1.27 to 2.73 pm, and average relative lengths ranging from 0.08 to 0.17 pm. No abnormalities were detected in any of the cells.
