Barrion Characteristic banding patterns of darkly stained regions separated by pale regions along the longitudinal lengths of the human mitotic chromosomes were observed after subjecting the chromosome preparations to modified C- and G- banding techniques. Karyotypes of these banding patterns were constructed according to Bregman (1990) utilizing photomicrographs of C-metaphase spread of C- banded (Category 1 chromosomes) and Pancreatin C-banded (Category 3 chromosomes) human mitotic chromosomes focused under phase-contrast microscope, interpretative drawings, chromosome lengths and band homology. Results showed that both banding techniques rely heavily on Giemsa to reveal AT-rich regions which are highly condensed during interphase. Specifically, C-bands are repetitive DNA sequences located near or on flanking centromeric regions of the chromosomes. G-bands, on the other hand, are moderately repetitive DNA sequences located in intercalary heterochromatic region of the chromosomes. Also, locally established C- and Pancreatin G- banding patterns of human mitotic chromosomes when compared to previously established C- and G-banding patterns revealed that constitutive heterochromatin located at the centromere were preferentially stained in C-banding while intercalary heterochromatin dispersed along the chromosome arms were stained in 0-banding. Discrepancies observed were due to different conditions of the chromosomes used and that overlapping and overcrowding of chromosomes affect banding patterns.
