ABSTRACT
Twenty-nine mangrove bacterial isolates collected from Pagbilao, Quezon, which were deposited at the National Institute of Biotechnology and Molecular Biology (BIOTECH) Environmental and Industrial Biotechnology Laboratory were screened and partially characterized for alginase activity. Five bacterial isolates were positive for the presence of the alginase enzyme. Quantification of the activities of the bacterial isolate was determined by a reducing assay using the Nelson-Somogyi method. The activities of the extracellular enzyme of the bacterial isolates were not significantly different from Vihrio harveyi, a known alginase-producing organism. When phenotypic characterization of the bacterial isolates was determined, cells of bacterial isolates 2p, 6a and 8a exhibited spherical shape while 10a and 12a were both rod shaped. Only isolate 6a had punctiform colony morphology while the rest had circular shape. All of the bacterial isolates were gram-negative and tested positive for the catalase test. When the alginase genes of the bacterial isolates were PCR-amplified, the amplicons of the isolates exhibited more than one alginase gene. V. harveyi had three amplicons, bacterial isolate 10a had four and isolate 6a had predicted two alginase genes. The optimization of pH, temperature and salt concentration was done for isolate 2p The optimum conditions for bacterial growth of 2p were pH 7, 7% NaCI and 37°C after 36 h of incubation.
