Abstract
The total muscle protein content of the freshwater prawn were determined using Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis (SDS-PAGE). A triangular collection, with three sites. Calamba (CAL), Victoria (VIC) in Laguna and Binangonan, Rizal (BIN), was used to determine variations in the protein content of the Macrobrachium lanceifrons Dana from different location within the lake A total of 29 bands were visualized in CAL and BIN samples, while only 28 bands were present in the VIC area There are 20 banding patterns in CAL 13 in VIC and 16 banding patterns were observed in BIN samples There are no similar banding patterns observed within the samples The collected species exhibited low SI values among samples and between sites This may be due to geographical distances since Binangonan found at the northern part of the lake and the other two sites are located at the southernmost tip Protein markers were used to approximate the molecular weight of major protein bands present in the samples. Four high molecular weight (HMW) protein markers and four low molecular weight markers were visualized The four HMW are thyroglobulin (330,000 D), albumin (67,000 D), catalase (60,000 D) and lactate dehydrogenase (36,000 D) The LMW markers visualized are phosphorylase b (94.000 D), albumin (67,000 D). ovalbumin (43.000 0), and carbonic anhydrase (30.000 D) Because of the similarities in their Rf values with that of some of the major protein bands, it was deduced that the °ands have the same molecular weights as that of the protein markers Band 1 has a molecular weight of 330,000 D. Band 17 (67,000 D). band 11 (94 000 D) band 19 (60,000 D), band 26 (36,000 D) and band 27 (30.000 D) All of these major bands are present in all samples of the three sites, except band 27 which is absent in CAL samples
