Biology major in Genetics. Adviser: Prof Celia B. Dela Viña ABSTRACT The appropriate stage of immature seed for the in vitro culture of pigeonpea, Cajanus cajan (L) Millsp. was determined using three accessions from Asian Vegetable Research and Development Center, namely TOT 0919, TOT 2492, and TOT 6099. Stage 1 seeds from 2.0- 2.9 cm long immature pods and Stage 2 from 3.0-3.9 cm long immature pods failed to grow in culture. Only Stage 3 seeds from 4.0-4.9 cm pods in all accessions that were inoculated on plain Murashige and Skoog (MS) medium survived. TOT 0919 exhibited the highest survival rate (70%), plant height (5.4 cm) and number of roots (7.8). On the other hand, TOT 2492, had the most number of leaves (3.7) and highest average root length (16.8 cm). Statistical analysis revealed that the interaction between the stages of immaturity, which was determined by pod length and accession, had a significant effect in all the parameters observed, at 5 % level. After transplantation, there were no significant difference in the growth and development of the three accessions indicating the adaptability of the cultured plant. With the failure of stage 1 and 2 seeds to grow in culture, seed viability assays were conducted. The germination and tetrazolium tests confirmed that the seeds were non-viable even before inoculation. Thus, in the germplasm collection of pigeonpea, immature seeds from 4.0-4.9 cm pods could be selected for in vitro culture.
