ABSTRACT

Short Sequence Repeat (SSR) markers that were newly developed for chromosome 6 in tomato were mapped in order to test their utility for genetic mapping. Previously, there were only seven SSR markers for chromosome 6. Selected polymorphic markers from this study were used to construct a new genetic map for chromosome 6. Molecular polymorphism between the F9 Recombinant Inbred Line population (RIL) from the cross Hawaii 7996 (Solanum lycopersicum L.) and WVa700 (S. pimpenellifolium) cross, and Backcross 1 (BC,) population from Hawaii 7996 x LA716 (S. pennellii), were assessed to check the segregation of the markers and generate linkage maps. Both the RIL and BC, mapping populations for tomato yielded a skewed segregation pattern favoring the Hawaii 7996 line. In the RIL mapping population, heterozygosity was observed despite the expected homozygosity in the mapping population. A total of 21 SSR markers that were mapped to chromosome 6 covered a total map distance of 94.5 cM with an average space distance of 4.97 cM. For the BC, population, 22 markers spanned a total map distance of 89.5 cM with an average map distance of 4.71 cM. This average distance is smaller compared to that of the R1L mapping population. When the two linkage maps were compared, 10 SSR markers were found to be present in both mapping populations but their order and map distances varied from one another. The gap, due to the absence of polymorphic markers between LE6-51 and LE6-53 in the linkage map of the RIL population, was bridged by the markers LE6- 22 and LE6-20 in the BC, population. The SSR markers developed were found to be efficient for genetic mapping of chromosome 6 in tomato.
