Abstract
lsozyme polymorphism in Christmas orchid Calanthe triplicate, (Willem.) Ames and Cleisastoma suhulatum Blume was investigated based on seven enzymes namely. shikimate dehydrogenase (SD1-1), alkaline phosphatase (ALPH). esterase (EST), malate dehydrogenase (MDH), acid phosphatase (ACPII). phosphoglucomutase (PGM), and glucose-6-phosphate dehydrogenase (G6PD) using starch gel electrophoresis. Isozyme banding patterns showed a total of nine presumptive loci. SDH, ALPFI, EST, MD1-1. and G6PD exhibited only one locus with genotypes SDHI. ALPHI. ESTI. ,VID111. and G6PDI while ACPH and PGM have two isoloci with genotypes ACP1-11. ACPH2. PG,111 and PGM2. The degree of genetic variability in the two Philippine orchids was determined by estimating average heterozygozity (H). proportion of polymorphic loci (P). average number of alleles per locus (A), genetic identity (Is). genetic distance (D). and genotypic similarity (1,i). The average heterozygozity, proportion of polymorphic loci. and average number of alleles per locus were all higher in C triplicate, (Willem.) Ames with values of 0.240, 62.50%. and 1.625. respectively as compared to those of C. suhulatum Blume with values of 0.111, 42.86%. and 1.429. respectively. High values of genetic identity (0.672) and genotypic identity (0.575) and low value of genetic distance (0.397) indicate that the two orchid species are somewhat genetically similar with each other based on the examined isozymes.
