Carica papaya latex (CPL) lipase, an sn-3 stereoselective enzyme with a protein content of 0.42 mg/mL and a specific activity of 16.07 Units/mg protein was used to catalyze the interesterification of coconut oil. The reaction conditions were optimized by varying the incubation temperature and time of the system. Hydrolysis and interesterification reactions that occurred simultaneously were monitored by HPLC analysis of the interesterified glycerides and by GLC of liberated FFA.
Changes in concentration of triglyceride were noted. The highest concentration of triglyceride formed in the product was 28.29%. This was obtained when the system was incubated at 65°C for 12 hours. The fatty acid profile of structured lipids and physical blend showed that there was a 37.17 % increase in the oleic and linoleic acid content of structured lipids compared to that of the physical blend. An increase or decrease of these fatty acids during the preparation of structured lipids indicate that interesterification took place.
