Antagonistic activity of bacteria isolated from local honey on Ascosphaera apis L., the causal organism of chalkbrood disease of honeybee

Abstract
Bacteria with antagonistic activity against Ascosphaera apis, the casual organism of Chalkbrood disease, were isolated from local honey samples. Seven honey samples tested were from the giant honeybee (Apis breviligula), honeybee (A. cerana) and from the European honeybee (A. mellifera). Thirty-one of the bacteria obtained were gram-positive, endospore forming rod while six were gram-negative. Six isolates from A. mellifera honey (MPA1, MPA2, MIS1, MTB6, and MTB9) and two from A. breviligula honey (BAB1 and BAB6) showed antagonistic activity against A. apis. Of these isolates, the three that had the numerically highest diameters of zones of inhibition (ZOI) than the positive control, Nystatin at 1000 units/mL (3.2 mm) were further tested. The largest ZOI was from MTB9 (6.6 cm) and the least was from MIS2 (1.4cm). Molecular identification of the three isolates with the numerically highest ZOI using 16s rDNA sequencing revealed that isolates MTB6 and MTB9 were under the genus Bacillus with various possible species. Isolate MIS1 on the other hand, had possible sequences similar to an uncultured bacterium or under the genus Enterobacter.
