Phylogenetic analysis of stingless bees (Trigona spp.) from Batac Ilocos Norte and Guinobatan, Albay based on cytochrome oxidase I gene

Abstract

The cytochrome oxidase I (COI) gene in stingless bees Trigona spp. collected from Ilocos Norte and Albay was characterized and used to assess genetic diversity. Out of seven published mitochondrial primer pairs, only mtD7 and COI-IIR consistently produced amplicons. The amplified DNA region was sequenced and BLASTSn results showed that the amplified region is highly related to Melipona bicolor COI gene. Raw forward and reverse complement sequences of COI gene were trimmed using PreGap 4, Trev 1.9 and Gap4 prior to analysis. Analysis of the COI gene sequence was carried out using Mega V5.2 with Bombus terrestris (bumble bee) as the outgroup. The samples from Albay clustered together with a bootstrap value of 98% in the Maximum likelihood Tree generated. This was further supported by the obtained pairwise evolutionary divergence values ranging from 0 - 0.019 indicating high genetic similarity among the colonies from Albay. The same was observed for the colonies from Ilocos with a value of 0.006 which indicated that there is likewise low divergence among the colonies. The rate of evolutions of Trigona spp. in Albay and in Ilocos Norte are equal based on the obtained P value which is less than the test statistic P=0.05. This indicates that the geographic situation of the two localities affects the rate of evolution of Trigona spp. inhabiting the area. Lastly, the computed mean interpopulational value is 2.147 indicating that the samples from Ilocos Norte are genetically distinct from the samples from Albay based on the COI gene. Amino acid sequences derived from the translated nucleotide sequences also showed high similarity to the COI protein of Melipona bicolor.
