Determination of phenolic content and antioxidant activity of Apis mellifera honeys in select regions of the Philippines

Abstract

The antioxidant activity of ten honey samples collected from select regions in the Philippines was determined by measuring the absorbance at 450 nm (ABS450), Folin-Ciocalteau, Aluminum chloride (AlCl3), diphenylpicrylhydrazyl (DPPH), and Ferric Reducing Ability of Plasma (FRAP) method to determine the intensity, Total Phenolic Content (TPC), Total Flavonoid Content (TFC), radical scavenging activity, and reducing ability of the samples, respectively. The darker honey was found to exhibit greater antioxidant activity. Floral source of each honey samples was determined by pollen analysis. The multifloral honey sample with dark amber color, composed of 40.96% Leguminosae II, 32.34% Palmae, 8.46% Myrtaceae, 6.63% Mimosa pudica, 5.97% Rubiaceae, 3.15% Cucurbitaceae, 1.16% Leguminosae III, 1% Portulaceae, 0.33% Euphorbiaceae was found to have the highest TPC and TFC with 946.77 ± 1.78 mGAE/kg honey and 247.56 ± 2.06 mCE/kg honey, respectively, among the samples with an intensity of 1246.11 ± 30.82 mAU. It was also found to exhibit the highest antioxidant activity with 7.63 ± 0.05 mg/mL (IC50) and 1061.74 ± 1.08 μmol Fe(II)/kg honey for DPPH and FRAP, respectively. The water white honey sample that was found to contain 84.39% Mimosa pudica, 13% Cucurbitaceae, 0.77% Mimosa diplotricha, 0.77% Graminae, 0.39% Palmae, 0.19% Euphorbiaceae exhibits lowest intensity (108.00 ± 0.67 mAU), TPC (67.61 ± 1.00 mGAE/kg honey) and TFC (101.57 ± 2.35 mCE/kg honey), and antioxidant activity (43.26 ± 0.37 mg/mL by DPPH and 171.51 ± 2.25 μmol Fe(II)/kg honey by FRAP).
