Abstract
This study is involved in the isolation and characterization of leptin gene in Philippine native pigs. It also aims to identify and find polymorphisms in the leptine gene of Philippine native pig. DNA sample used in this study came from the ear tissues of Philippine native pig collected from the Animal and Dairy Science Cluster farm. The obtained sequences were analyzed using the BLAST n search for sequences from the database that were homologous to the sample sequence. The ADSC native pig sequence has a 98% homology with leptin gene sequences in pigs. Multiple sequence alignment of the sample sequence and other pig leptin sequences was performed and showed a 94.6% homology. Nine single nucleotide polymorphisms were identified. To further analyze the native pig sample sequence, protein sequence alignment was done. One change in the amino acid sequence was observed. The sequences showed 99.2% homology. Detailed study on the polymorphisms and its relation with growth, reproduction, and meat quality of Philippine native pigs is recommended. It is recommended that more samples of Philippine native pigs from different regions be collected and be cloned to ensure purification and good quality of sample for sequencing.
