DNA sequence analysis of the heat-shock protein 70 gene promoter of selected Philippine native chickens (Gallus gallus Linnaeus)

Abstract

Abstract: Philippine native chickens are known to be heat resistant. Studying this can become very helpful for breeding. There are a lot of studies about heat resistance in chickens, but the heat resistance of the Philippine native chickens has not yet been studied, especially in the molecular level. During heat stress, the Heat Shock Protein 70 (HSP70) prevents cellular damage. Its promoter is one of the main factors that affect its regulation. We compared the sequence of the promoter of different Philippine Native Chicken breeds and with the promoter sequences of other chicken breeds available in the NCBI database. DNA were extracted from the blood samples of three Labuyo, three Darag and two Cobra chickens using FTA® cards. PCR primers were designed to target the promoter region of HSP70 and optimized. HSP70 promoter regions of Labuyo, Darag and Cobra were PCR amplified and sequenced. Sequencing results showed that there were no polymorphism in the HSP 70 gene promoter region sequences within and between the chicken samples analyzed. However, when the samples were compared with the HSP70 gene promoter sequences from the NCBI database, polymorphism was evident in the 8th, 301th, 318th and 353th to 355th base pairs from the 5? end of the promoter sequence. A 98.6% homology was observed between samples and the Kadaknath, Rhode Island Red and White Leghorn sequences. Similarly, the said samples showed 98.4% homology with the White Plymouth Rock. With these findings, further studies should be conducted using other breeds of chicken.
