Abstract
The effect of varying substrate concentrations specifically glucose, molasses, and coco-paring meal of the basl medium on the biomass concentration and emulsification activity of the biosurfactant produced by Saccharomyces cerevisiae 2030 was analyzed through shake flask system. Al,of the data subjected to one-way Analysis of Variance (ANOVA) using Duncan's Multiple Range Test (DMRT) at 5% confidence level. The preferred condition to favor growth of microorganism and increase emulsification activity of the biosurfactant was set at minimal concentrations at 2% (v/v) molasses, 2% (v/v) glucose and 2% (v.v) coco-paring meal because of the insignificant effect of increased values. The bench sale production resulted in about 80% emulsification. Effects of some factors such as pH and fermentation time also analyzed in this study. The highest emulsification index was obtained at pH 8. In terms of fermentation time, there was no significant effect on emulsification activity on day 3 to 7. Some components of the biosurfactant were also characterized such as carbohydrate and protein employing phenol sulfuric acid assay and method of Lowry, respectively. The biosaurfactant showed no surface tension reduction activity using the modified drop collapse method.
