Abstract
Evaluation of the ethanol fermentation performance of the acid tolerant yeast strain Saccharomyces cerevisiae 5N1 was done using 2ᵏ full factorial experimental design for the 16 flask fermentation runs in order to determine the effect of pH and Leuconostoc mesenteroides contamination with S. cerevisiae HBY3 as the bench mark strain. Results of Analysis of Variance and Duncan's Multiple Range Test showed that bacterial contamination (10E+04) cells/mL) had no significant effect on the measures of ethanol fermentation performance suggesting effective management by lowering the initial pH of the molasses medium up to pH 3.0. On the other hand, pH had a significant effect on the hand. pH had a significant effect on the amount of amount of ethanol produced, fermentation efficiency , volumetric ethanol productivity as well as on other parameters like CO₂ evolution, sugar consumption and biomass and cell concentration. Better results were obtained at pH 4.5 than at pH 3.0 but strain 5N1 still got better results even at lower pH due to its acid-tolerance. All fermentation results proved its remarkable potential as an alternative strain for industrial scale fermentation for its highest attained value of ethanol produced which was 11.53% (v/v) or 90.97g/L, overall fermentation efficiency of 88.91% and a very high volumetric ethanol productivity of 2.74 g ethanol/L-hr even after only 24 hours.
