Effect of equilibration time on the post-thaw motility of buffalo spermatozoa frozen using different concentrations of ethylene glycol

Abstract
This study was conducted to determine effects of equilibration time (20, 30, 40, and 50 min) on the post-thaw motility of buffalo spermatozoa using different concentrations (7, 8, and 9%) of ethylene glycol. Six bulls were used for semen sampling, and the semen was diluted with Tris-based extender, and sealed. The straws were placed at 4 cm above the level of nitrogen for seven minutes, and then submerged in liquid nitrogen. After at least one week storage, the samples were thawed and evaluated. Results of the study showed that the highest post-thaw motility rate (14.29%) was obtained with 7% ethylene glycol at equilibration time of 30 minutes. Results further showed that as the concentration increased to 9%, a corresponding decreased in the post-thaw motility was observed in various equilibration times. In conclusion, the study indicates that 7% of ethylene glycol is the optimum level for freezing with short equilibration time. However, obtained post-thaw motility rate is quite low. Thus, there is a need to study other factors that affect semen viability.
