Abstract
Seven rice varieties, namely, Dular, Fujisaka 5, IR 29, Makiling, Peta, PTB 21, and Vary Kongo, and a check (Taipei 309) were compared fort their embryogenic cell suspension ability as measured by their callus forming ability, embryogenic cellproduction, and plant regenerability. Callus formation was assessed based on the number of days to caalus formation and percent callus induction. IR 29, Makiling, and Peta were found to form calli as early as 4 days after inoculation. Fujisaka 5, however, took the longest time to form calli at 10 days. Frequency of callus induction was not significantly different among the differences varieties. Embryogenic cell production, on the other hand, was evaluated based on weight increment of calli, degree of browning and percent embryogenic cell suspension easily as supported by the cytological evaluation which showed that its calli were composed mainly of embryogenic cells. Of the other seven varieties, Dular was the only variety comparable to Taipei 309 in terms of the frequency of embryogenic cells as evaluated by the cytological evaluation. Other varieties showed ample amounts of embryogenic cells but they were not able to establish lines. This might be attributed to the selective action of the media. Furthermore, only Taipei 309, Makiling and Peta were able to regenerate.
Thus, none among the different varieties tested were comparable to Taipei 309 in over-all embryogenic cell suspension ability. Certain modifications in the procedure must be undertaken to have a more desirable response to embryogenic cell production.
