Abstract
A study was conducted to determine the relative case of establishing Calli from adapted and non-adapted wheat cultivars, evaluate the effect of various short-term heat treatments (i.e. temperature and length of exposures) on growth of E-callus and regenerants from adapted and non-adapted wheat cultivars and assess subsequent plant generation from such heat pretreated calli.
Two adapted cultivars, Trigo 2 and Trigo 3 and two non-adopted cultivars, Siette Cerros and Seri 82 were assessed for their capacities to induce calli, the number of days to callus induction, the type of callus formed and the frequency of callus types. The incidence of callus formation did not vary significantly for the different cultivars. However, differences in incidence of callus induction at 2 or 3 days, callus origin and incidence of callus types were significant among adapted and non-adapted wheat cultivars.
Embryogenic calli that were used to evaluate the effect of short-term heat treatments on adapted and non-adapted cultivars. Adapted cultivars gave a higher percentages of healthy callus and E-callus regrowth as well as faster increment in callus size compared to non-adapted cultivars. Subsequent shoot regeneration was higher for adapted cultivars than non-adapted cultivars due to greater E-callus survival from heat treatments.
Regenerants subjected to heat treatments did not significantly differ, thus, heat treatments at the callus level rather than plant level maybe a better approach for in vitro screening for elevated temperature.
