Isolation, characterization and identification of bacteria from lahar
Abstract
Samples of lahar and soil from Pampanga were collected and microbial counts were determined. In unaffected areas, the average colony-forming unit per milliliter (cfu) was 1.03 x 107 which is higher compared to that from the lahar-affected area (3.82 x 106 cfu/ml). Twenty-five of the 89 isolates were subjected to different morphological, cultural and physiological tests for their identification. Results of the tests showed that isolates from the surface of lahar-affected area belong to the following genera and species: Plesiomonas shigelloides (isolate L1-1), Acetobacter hansenii (L1-2), Acinetobacter calcoaceticus (L1-3), Proteus mirabilis (L1-4), Aeromonas caviae (L1-S), Bacillus coagulons (L1-6), Citrobacter freundii (L1-7), Kurthia gibsonii (L1-8), Micrococcus agilis (L1-9), and Bacillus subtilis (L1-26). For samples obtained from 5 cm depth in lahar-affected area, the following were identified: Micrococcus varians (isolate L2-1), Aeromonas caviae (L2-3), Staphylococcus (L2-4), Kurthia gibsonii (L2-5), Bacillus laterosporus (L2-6), Plesiomonas shigelloides (L2-7), Micrococcus agilis (isolates L2-8 and L2-11), Legionella morganii (L2-9) and Cellulomonas biazotea (L2-10). Different test results for isolates from 10 cm depth in lahar-affected area identified Micrococcus agilis (L3-1). In unaffected area, isolate UL1-1 obtained from the surface was identified to be Bacillus laterosporus. From 5 cm depth samples, different test results for isolate UL2-7 conformed with the characteristics of Bacillus laterosporus. Isolate UL3-3 and UL3-5 from 10 cm deep samples were identified to be Kurthia gibsonii based on their different test results.
