Genetic mapping of isozyme markers in mungbean (Vigna radiata L. Wilczek) and blackgram (Vigna mungo L. Hepper). I. Standardization of electrophoretic procedures and survey of marker polymorphism
Abstract
A preliminary screening of isozyme polymorphism in mungbean (Vigna radiata L. Wilczek) and closely related Vigna species (V. radiata ssp. sublobata and V. mungo L. Hepper) was carried out in view of the use of the data for genetic mapping in mungbean. Isozyme analysis was conducted using dry seeds and young leaf. Starch gel and polyacrylamide gel electrophoresis of tissue extracts resolved at least 15 reproducible banding patterns out of the 24 enzyme systems tested. From the 24 enzyme systems surveyed among the parental lines, 10 seed enzymes and 8 leaf enzymes were found polymorphic. Greater polymorphism was observed between the parental pair, V. radiata and V. mungo, compared to V. radiata and V. r. sublobata.
