Effect of chemical and physical treatments on the in vitro germination and growth of 'Laguna Tall' coconut embryos

Abstract

In vitro studies involving 'Laguna Tall' coconut embryos were conducted to assess the germination and growth of seedlings in response to different chemicals added to the Y3 medium, manner of culture and different light spectra treatments during incubation. Supplementation of gibberellic acid at 30 and 40 μM into the Y3 medium significantly promoted embryo germination and shoot emergence of seedlings, but at a higher concentration of 50 μM, germination was significantly inhibited. Similarly, supplementation of 10-40 μM GA₃ promoted shoot elongation of germinated coconut seedlings, but 50 μM GA₃ inhibited shoot elongation. Abscisic acid at 0.94-3.78 μM added to the Y3 medium also promoted germination of embryos, but higher concentration of 7.67 μM significantly decreased germination. However, silver thiosulfate, spermidine and the state of culture had no effect on embryo germination and growth of seedlings. The different visible light spectra i.e. blue (430-500 nm), green (500-560 nm), yellow (560-600 nm) and red (650-750 nm) significantly affected fresh weight and shoot length of germinated seedlings 16 weeks after explanting. Incubation of cultures under red light spectrum (650-750 nm) significantly increased the fresh weight of coconut seedlings compared to those seedlings incubated under white fluorescent light.
