Laboratory efficacy test of transgenic eggplant with Bt cry1Ac protein against the Los Baños population of eggplant fruit and shoot borer, leucinodes orbonalis Guenee (Lepidoptera: Pyralidae)

Abstract

Feeding experiments using detached tissue samples from BC3F1 populations of eggplant crosses Mara x MAHYCO event EE-1 and DLP x MAHYCO event EE-1 consisting of 10 BC3F1 lines and 9 BC3F1 lines, respectively, were conducted in the Institute of Plant Breeding (IPB) Entomology P2 Laboratory to determine the level of efficacy against the Los Baños population of eggplant fruit and shoot borer (EFSB), leucinodes orbonalis Guenee (Lepidoptera: Pyralidae). Terminal leaves and immature fruits (about two weeks old) were collected from Bt positive and Bt negative plants of BC3F1 Mara x MAHYCO EE-1 and BC3F1 DLP x MAHYCO EE-1. The samples were from plants randomly selected across each replication of eggplant lines planted in the IPB confined trial site. The detached leaf and immature fruit bioassays were done using neonates of laboratory-reared EFSB. The cumulative percent mortality of EFSB larvae using the two assay tests were recorded at 0.5 to 48 hours after release on detached leaves, and 3 to 8 days after release on immature fruits. The results showed high level of efficacy of the EE-1 event against the Los Baños population of EFSB. A 100% mortality of EFSB larvae was observed on the detached leaf bioassay after 48 hours of feeding the EFSB with terminal leaf of Bt positive Mara x MAHYCO EE-1 and DLP x MAHYCO EE-1 backcross plants, compared with only 10-11% mortality when fed with leaves from the Bt negative plants. In addition, results of the detached immature fruit bioassay showed that fruits from Bt positive plants of event EE-1 effected 100% mortality of EFSB neonates within 6-8 days. compared to just 24% and 30% of those fed with fruits from the Bt negative BC3F1 Mara x MAHYCO EE-1 and BC3F1 DLP x MAHYCO EE-1 plants, respectively, during the same observation period. These observations indicate that growing transgenic Bt eggplant is an effective strategy for the control and management of EFSB.
