Genetic diversity analysis of twenty three CLSU special rice variety collections

Abstract

Twenty three rice varieties from CLSU [Central Luzon State University] collection and three check varieties from PhilRice [Philippine Rice Research Institute] were used in morphological characterization for 44 traits, grain quality analysis for six parameters, molecular characterization for 63 SSR markers, and for market fragrance genotyping. This study aimed to: a) determine the diversity (genetic and morphological) of 23 aromatic rice accessions of CLSU; b) identify entries that could be used as parents in the production of variants in the breeding program of CLSU; and c) validate each entry as an aromatic rice variety by means of molecular and aroma analysis. There were nine cluster made by morphological characterization, and six clusters with two sub clusters made by molecular characterization through SSRs. Morphological clustering was distinguished by unique traits in most clusters (I, II, VI, VII, VIII, and IX) and also by combination of different traits may be present scattered by with other non member entries in some clusters (III and IV). Molecular characterization distinguished clusters into indica type, basmati type, jasmine type, Pakistan origin, Leyte origin, distinct traditional variety, and temperate japonica. Moreover, 15 entries were confirmed aromatic by sensory analysis and marker fragrance genotyping for 2-acetyl-1-pyroline. The entry IR841-14-2 was analyzed as superior variety in grain quality analysis. The study showed that there is wide genetic diversity among accessions used. The avoidance of selecting two accessions within the groupings from the study can guide the breeders in crossing more diverse parents that may result for better variability in their progenies. Moreover, the validation for aromatic character within the collection can help the breeders to create aromatic offsprings since the character for aroma is known to a recessive trait and requires homozygosity for expression.
