Abstract
This research internship focused on the genetic diversity of Rhizoctonia solani causing rice sheath blight in the Philippines. The research internship was conducted at the Plant Breeding, Genetics and Biotechnology Division, Plant Pathology laboratory in D.L Umali Building, International Rice Research Institute from June 2013 to March 2014. The laboratory activities involved collection of samples from different geographic locations mainly in Luzon, isolation of isolates into pure culture, culture preservation, DNA extraction of R. solani isolates, and use of the software DendroUPGMA and GenAlEx to analyze the relationships between and among 94 isolates of R.solani from the molecular genotyping generated at the Swiss Federal Institute of Technology (ETH) Zurich. Some improvements were recommended to overcome the problems during the DNA extraction of the pathogen. Geographical grouping was observed only from Leyte and some isolates from Laguna, Nueva Ecija and Pangasinan. Genotypic genetic distance was highest between Ilocos and Nueva Ecija (11.964) and lowest between Leyte and Laguna (8.465), which coincide with the output generated from DendroUPGMA.The chi-square tests for Hardy-Weinberg Equilibrium showed that all populations except Ilocos were in HWE suggesting the importance of sexual reproduction of R. solani in the field. The importance of molecular tools in diversity studies was highlighted in the internship in order to formulate better management strategies against the disease.
