Abstract
The genetic variability of 35 Ralstonia solanacearum (E. F. Smith) Yabuuchi et al. isolates obtained from soil planted to different solanaceous crops in Benguet, Misamis Oriental and Bukidnon was analyzed. All isolates were confirmed as Ralstonia solanacearum based on their colony characteristics on Tetrazolium Chloride Agar (TZCA), generation of the 281-bp amplicon by PCR using species-specific primers 759/760 and their pathogenicity to tomato. Multiplex-PCR using phylotype-specific primers revealed that Mindanao isolates belong to phylotype I while the Benguet isolates belong to phylotype II. Repetitive element (rep) – PCR using REP and BOX primers was employed to assess the genetic diversity of the R. solanacearum isolates. Eight and 13 haplotypes were defined by REP and BOX, respectively. Cluster analysis by UPGMA grouped both the REP and the BOX haplotypes into two big clusters at 50% similarity level. The Mindanao isolates formed one cluster and the Benguet isolates formed another cluster. The separation of the Benguet and the Mindanao isolates into two clusters revealed the differentiation of the isolates according to geographical origin. More clusters were formed at 70% similarity level. However, the separation of the Mindanao from the Benguet isolates was still evident. High haplotypic diversity estimates of 0.83 and 0.82 were obtained with REP and BOX, respectively. This suggests that there is a high level of diversity among the isolates of Ralstonia solanacearum that were analyzed.
