Abstract

Caseins are proteins in milk that exist in various forms (alpha S1, alpha S2, beta, and kappa) with variant alleles of each. Among the variants of beta-casein, variants A1 and A2 are the most common in major native cattle breeds. Beta-casein variant A2 is reported to be favorable for milk quality and is considered to be included in breeding strategies of dairy animals. The main objective of this study was to genotype the top 25 animals from a population of 100 Siquijor native cattle for the beta-casein gene (CSN2) by polymerase chain reaction sequencing method. A 650-base-pair (bp) CSN2 fragment was amplified using primers Rp.7i (5' GAC AAT CAT TAA TCT CAG CTA TGC 3') and Fp.7c (5' GAT TTG TTT TCC TTC TTT CGA G 3′). Results showed that among the genotyped Siquijor native cattle (25), 15 were found to be of genotype A1A1, and 8 were of A1A2 genotype. The variant A1 has histidine in the 67th amino acid of the chain that causes to produce BCM-7 that leads to various diseases, while the variant A2 does not. Two (2) of the cattle in this study were homozygous for beta-casein genotype A2A2, and it is concluded that beta-casein can be used as a DNA marker in bovine selection program for production of milk with high quality and that the Siquijor native cattle can be used as a foundation stock for dairy cattle.
