Morphometrics and amplicon sequencing of 16S rRNA genes of stingless bee (Tetragonula sp.)

Abstract

Morphological characterization and gene sequencing of two colonies of stingless bees obtained from Sorsogon, Bicol region were carried out. Quantitative-qualitative methods were used in describing the morphology while genomic DNA extraction, polymerase chain reaction and agarose gel electrophoresis were done at the Philippine Genomic Institute, University of the Philippines Diliman. Results were outsourced at the 1st BASE DNA Sequencing Service for single-pass capillary sequencing and analyzed using the Basic Local Alignment Search Tool (BLAST). Morphology of the egg, larva, pupa, and adults (worker and queen) were differentiated in terms of the color and sizes. BLAST analysis revealed that the two samples morphologically confirmed as T. biroi had only amplicons from 16S rRNA genes reproducible on the colonies analyzed. The searches showed that the forward and reverse sequences have 92% identity with those of the old world stingless bee, Tetragonula pagdeni and 90-91% identity with the Sumatran T. collina, CS-1 and CS-2 samples gene sequence is for T. biroi, a new data base for further sequencing of the whole genome.
